Moreover, the controlling the splicing efficiency can quantitatively regulates the suppression effect on RNAi.胚乳特異的な遺伝子機能の破壊及びRNAiの量的調節について解明するため、胚乳特異的プロモーターで誘導され、第一イントロンをループに持つRNAiベクター、pWRI-A、pWRI-Bを構築した。pWRI-Bは第一イントロンの5'側スプライシング部位に塩基置換があり、RNAi特異的二本鎖RNAの蓄積量が減少する。両方のRNAiベクターとも96~100%の形質転換体で内在Wx mRNAの発現抑制が観察された。さらに、WRI-B/Wxbの抑制効果はWRI-A/Wxbよりも弱かった。これらの結果により、Wxプロモーターが胚乳特異的なRNAiを誘導させることが明らかとなった。さらに、スプライス効率の調節がRNAiの抑制効果を制御できる可能性が示唆された。 | |||||
