It is widely accepted most tumor cells display increased glucose uptake and glucose metabolism compared to normal cells. Vasculature of brain neoplasm is characterized by abnormal properties important to tumor biology and therapy. This investigation elucidates the association between morphological properties of vasculature, vascularity, permeability, and gene expression of glucose transporter (GLUT) of brain tumor. In the tumor, three regions can be identified on the basis of vascularity and histology: peripheral, intermediate, and central zone. HRP extensively extravasated from abnormally dilated tumor vessels, densely filling the widened extracellular space (ECS) in the intermediate zone. Northern blotting analysis indicated GLUT3 mRNA were up-regulated in each tumor. In situ hybridization revealed that GLUT3 mRNA expression was markedly increased in tumor cells facing necrosis. Increased permeability and widened ECS may provide efficiency in obtaining substrates for energy and cellular proliferation of the tumor. 内容記述 Glucose is the major substrate for brain tumor energy metabolism. It is widely accepted most tumor cells display increased glucose uptake and glucose metabolism compared to normal cells. Vasculature of brain neoplasm is characterized by abnormal properties important to tumor biology and therapy. This investigation elucidates the association between morphological properties of vasculature, vascularity, permeability, and gene expression of glucose transporter (GLUT) of brain tumor. In the tumor, three regions can be identified on the basis of vascularity and histology: peripheral, intermediate, and central zone. HRP extensively extravasated from abnormally dilated tumor vessels, densely filling the widened extracellular space (ECS) in the intermediate zone.
