ADの最大の遺伝的リスクとしてAPOE遺伝子多型が知られている. Aβは, その代謝システムとしてアポリポ蛋白E(ApoE)と複合体を形成し細胞内に取り込まれる機序が知られている. LR11/SorLAはAβの産生・代謝を調節するリポ蛋白受容体であり, その細胞外領域にApoEと結合するドメインを有している. 本研究では, 全長型LR11(full-length LR11:FL-LR11)がisoform依存性にApoEを細胞内に取り込む可能性について培養細胞を用いて検討した. HEK293T細胞にApoE-GFPを遺伝子導入し, その培養液をFL-LR11を導入したN2a細胞に添加し, 分泌ApoE-GFPのN2a細胞への取り込みを検討した. N2a細胞を共焦点レーザー顕微鏡で観察すると, 培養液中のApoE-GFPがLR11を介して細胞内に取り込まれた. ApoE2, E3, E4を遺伝子導入したドナーHEK293T細胞とFL-LR11を遺伝子導入したレシピエントN2a細胞の共培養実験を行ったところ, LR11の強制発現により分泌ApoE3及びApoE4のレシピエントN2a細胞内への取り込みが亢進した.一方で, 分泌ApoE2ではレシピエントN2a細胞への取り込みの亢進は認めなかった. 共免疫沈降解析を行ったところ, ApoEとLR11は細胞の内外で複合体を形成した.
