A 1.8 Kb XbaI fragment was isolated from two different libraries and its sequence determined by the dideoxy and chemical cleavage methods. The H-2D^p XbaI fragment consists of 1,837 nucleotides and includes the 5'-flanking region, the L, N, and Cl domain coding regions and intron 1, 2 and 3. The H-2D^d clone is of similar length and includes comparable regions of the gene. The D^d sequence, with the exception of three nucleotide substitutions, was identical to the published sequence of pAG64, a cDNA clone obtained from a SV40 transformed 3T3 cell line (5). Comparison of the nucleotide sequences of the D^p and D^d gene fragment reveals a high degree of homology (92%). Further comparisons of the D^p and D^d sequences with other previously published classI gene sequences reveals that the homology between D^p and D^d is higher than the homology of either of these genes to D^b, L^d, K^b or K^d. The small differences in the 5'-portion of D^p and D^d gene are up to three nucleotides.
