The gel fractionation proved to be reliable in collecting platelets free from plasma. Collected platelets were centrifuged with ACD, incubated with ammonium oxalate, centrifuged and resuspended in phosphate buffered saline as substrate platelets. Reactions between the substrate platelets obtained from blood group-0 healthy men and alloimmunized anti-platelet antibodies, i.e., anti-Yuk^a and anti-Yuk^b serum, were analyzed by a laser flow cytometer, Cytoron (Ortho Diagnostic Systems), with satisfying results.
