Highly enriched γδ-receptor bearing T cells were prepared from the peripheral blood mononuclear cells (PBMC) of three healthy donors and six patients with malignant glioma. After culturing with phytohemagglutinin and recombinant interleukin (rIL)- 2, 46-92% of PBMC came to bear the γδ T cell receptor, which hereafter shall be referred to as gamma delta riched (GDR) cells. GDR cells from one of the healthy donors showed a wide ranging killer activity against glioma cell lines as well as Raji cells. GDR cells which lysed autologous tumor cells also showed strong cytotoxic activities against the allogeneic glioma cell line U251 in a nonmajor histocompatibility complex restricted manner.
