[^<18>F]2-FDG(2-fluoro-2-deoxy-D-glucose)を用いたPositron Emission Tomography (PET)が確立されているが, 放射線被曝に対する厳密な対応の必要性から大掛かりな施設が必要であり, 測定コストも大きい. 磁気共鳴イメージング(MRI)は放射線を用いず,非侵襲的に生体の形態・機能を見ることができる優れた画像法である. 今回, 糖代謝イメージングを目的として, 3-FDG(3-fluoro-3-deoxy-D-glucose)を用いた^<19>F MRIを開発, 正常マウスとアルツハイマー病遺伝子改変マウスに応用した.【方法】実験は7T実験用磁気共鳴装置を使用し, ^<19>F 3-FDGファントムを用いて,パルスシークエンスの作成・調整を行った. この測定法を用い, 3-FDG(500 mg/kg)をC57BL/6正常マウスに経静脈的投与し, ^<19>F MRIの測定をおこなった. 得られたraw dataはMatlab上で自作のプログラムを用いて処理し,イメージ処理を最適化した. ついで, 7ヶ月齢雄のアルツハイマー病遺伝子改変マウス(5XFAD)と正常マウス(C57BL/6)(各n=5)に対して^<19>F 3-FDG MRIを施行, β3-FDGを目的分子としたイメージングの関心領域の解析を行い,大脳皮質, 基底核, 嗅脳の対小脳比を算出し比較検討を行った.更に両マウス(各n=5)に対して^<19>F 3-FDG MR spectroscopic imaging (MRSI)にて, 3-FDGおよびその代謝物(3-FD sorbitol)を同時測定し, 3-FDGの代謝経路としてのポリオール経路の酵素(Aldose reductase)活性を比較した.【結果】バンド幅420HzのGaussian pulseでβ3-FDGの選択的励起をおこない, hyperbolic secant pulseによりスライス選択し, echo signalの保持にはBIR4 adiabatic pulseによるmulti echo採取をおこなうadiabatic fast spin echo法を開発し測定に用いた. 正常マウスにおけるイメージ最適化には, Gaussian kernelをk-spaceに乗しzero fillingを行ったのちに離散フーリエ変換を行うことでS/N比と空間分解能の向上を得た. 5XFADマウスと正常マウスとの^<19>F 3-FDG MRIにおける対小脳比の相対信号の比較では, 5XFADマウスの大脳皮質で正常マウスよりも有意に高値であった. ^<19>F 3-FDG MRSIでは,Aldose reductase活性を示すSorbitol indexに5XFADマウスと正常マウス間で有意差は認めなかった.【結論】3-FDGを用いた^<19>F MRI adiabatic fast spin echo法を脳糖代謝イメージングとしてマウス脳に応用した. ^<19>F 3-FDGMRIで, 7ヶ月齢の5XFADマウスの大脳皮質では, 相対的にβ3-FDGの取り込みが上昇していることが示された.この時,^<19>F 3-FDG MRSIではポリオール経路の亢進は認められなかった. 以上よりβ3-FDGの取り込み上昇は,
