syndrome，MetS)は，内臓脂肪型肥満を基盤として高血糖，脂質異常，高血圧が引き起こされる病態であるが，慢性腎臓病とも密接に関連することが示されている．しかし，MetSの腎，特に近位尿細管機能に関わる因子とその異常に関してはまだ十分に明らかにされてはいない．そこで，6週齢時に，MetSモデルであるOLETF(Otsuka Long-Evans Tokushima Fatty)ラットを食餌自由摂取群(OLETF)と，食餌制限群(pair-fed OLETF)にわけ，対照ラットとともに12週齢で評価を行った．OLETF群では対照に比較して体重，糸球体径，腎重量が増加した．さらに近位尿細管エンドサイトーシス受容体メガリンのリガンドであるアルブミン，β2-ミクログロブリン，ビタミンD結合蛋白(DBP) などの尿中排泄量も増加したが，食餌制限群ではどれも増加を認めなかった．また，腎内中性脂肪もOLETF群で増加したが，食餌制限群においては増加を認めなかった．腎皮質ではmonocyte chemoattractant protein-1(MCP-1)のmRNAがOLETF群で発現が亢進したが，食餌制限では亢進を認めなかった．また，MCP-1の受容体であるケモカイン受容体2 (chemokine receptor type 2，CCR2)の免疫染色では，CCR2がOLETF群の近位尿細管(S1領域)で発現が増加したが，食餌制限では増加しなかった．これらの結果から，MetS腎症ではメガリンの機能異常に代表される近位尿細管細胞の形質変化が起こり，そこには脂肪毒性やMCP-1-CCR2系が関与することが示唆された．","subitem_description_type":"Abstract"}]},"item_6_description_5":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"学位の種類:
