In this study, we tried to elucidate the usage of TCR β chain variable regions (Vβ) of WT1 or CMVpp65 specific CTLs by using TCR Vβ gene family-specific monoclonal antibodies and flow cytometric analysis. In pre-culture analysis, the usage of TCR Vβ was diverse in CD8+ T cells of all samples. Predominant samples from MLPC with high proportion of CD8+MHC tetramer+ cells within CD8+ T cells showed the usage of a single TCR Vβ gene. However, oligoclonal usage of the TCR Vβ genes was observed in cells from some wells of MLPC containing CD8+MHC tetramer+ cells. The type of TCR Vβ repertoire in MLPC cells from each well was not identical and showed oligoclonal feature from well to well.
