As methanol concentration (±20%) in eluent (0.5 mL/min) was changed using octadecylsilane (ODS) type column in 25 cm length, retention time became stable after 15 min. As to 6 standards of plant hormones, the relationship between methanol concentration in eluent and retention time was examined in each column. In ODS type column, the general tendency of elution pattern was confirmed. As two columns were connected, elution pattern was same as that of one column and no problem occurred. As in the elution pattern both MeOH concentration and retention time were plotted in log scale, extent of curve was gentle and the tendency of relationship between the two was confirmed. As injected indole-acetic acid (IAA) amount (solute amount) and peak area were plotted. correlation was high and there was proportional relationship. Regardless of injected IAA amount, as amount of injected MeOH was constant, value of half-band width was constant. As half-band width and MeOH concentration of eluent were plotted in log scale, they had almost straight relationship.
