{"created":"2021-03-01T06:34:18.071974+00:00","id":28484,"links":{},"metadata":{"_buckets":{"deposit":"559f6fec-e2ec-4ffb-9143-17e9c653fdf1"},"_deposit":{"id":"28484","owners":[],"pid":{"revision_id":0,"type":"depid","value":"28484"},"status":"published"},"_oai":{"id":"oai:niigata-u.repo.nii.ac.jp:00028484","sets":["453:456","485:872:1583:1608"]},"item_7_alternative_title_1":{"attribute_name":"その他のタイトル","attribute_value_mlt":[{"subitem_alternative_title":"Effect of Medium Composition on Soybean Seed Storage Protein, βSubunit of β Conglycinin, mRNA Accumulation in In Vitro Immature Cotyledon Culture System"}]},"item_7_biblio_info_6":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2004-08","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"1","bibliographicPageEnd":"45","bibliographicPageStart":"41","bibliographicVolumeNumber":"57","bibliographic_titles":[{"bibliographic_title":"新潟大学農学部研究報告"},{"bibliographic_title":"新潟大学農学部研究報告","bibliographic_titleLang":"en"}]}]},"item_7_description_4":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"ダイズ種子貯蔵タンパク質の一つであるβコングリシニンのβサブユニットの集積調節物質を検索するために、未熟子葉の試験管培養系を用いていくつかのアミノ酸、植物ホルモン及び有機酸添加に対する応答性を調査した。グルタミン酸、アラニンを添加すると、未熟子葉内の遊離アミノ酸が上昇し、いくつかのアミノ酸が増加したことから、これらのアミノ酸は種子に取り込まれ、代謝されたことが示唆された。しかし、βサブユニットmRNAはグルタミン添加培地以外では検出できなかった。アスパラギン酸添加では未熟子葉内の遊離アミノ酸濃度の上昇及びβサブユニットmRNAの集積は認められなかった。植物ホルモンの一つサイトカイニン(t-zeatin)の添加によってもβサブユニットの集積は促進されなかった。TCA回路のα-ケトグルタル酸、オキサロ酢酸、クエン酸の添加によってもβサブユニットmRNAの集積は促進されなかった。また培養後の未熟子葉内におけるこれらの有機酸濃度にはグルタミンのみ添加の対照区と比較して有意な上昇は認められなかった。以上の実験結果から、ダイズ未熟子葉において少なくとも今回試験に用いた物質の子葉外濃度の上昇によりβサブユニットmRNAの集積は促進されないことが確認された。It has been reported that the accumulation of soybean (Glycine max L.) storage protein, β subunit of β conglycinin, was suppressed by nitrogen (N)-deficiency. In this report we tried to evaluate what compound(s) may have a key role in regulating the accumulation of β subunit mRNA in vitro cotyledon culture system. The cotyledons from non-nodulating soybean plants cultivated under N-deficient condition (0.5 mM NO 3 -) were transferred to in vitro culture system. The application of glutamine (Gln) enhanced β subunit mRNA accumulation within twelve hours. The application of glutamic acid, alanine, or aspertic acid could not induced β subunit mRNA accumulation. Application of cytokinin (t-zeatin) oxaloasetate, 2 -oxoglutarate and citrate could not enhance the accumulation of β subunit mRNA under N free culture medium.
