Based on this observation, a new idea has been strongly proposed that the cytoplasmic matrix is structured and consists of two phases, i.e., a protein-rich phase represented by the microtrabeculae and a water-rich phase by the intertrabecular space. Since then, this fascinating idea has been exposed to serious criticism. The main notion against the idea is that the microtrabeculae are the artifacts derived from the procedures employed for the specimen preparation including the chemical fixation and the critical point-drying. The present author has presented evidence strongly suggesting that there are no meaningful differences in ultrastructural images between the chemically fixed specimens and the rapid freezing ones without the chemical fixation. However, he has also shown that the bovine serum albumin solutions exhibit lattices not unlike the microtrabeculae in the embedment-free section. Therefore, it is cautious to conclude simply that all the microtrabecular strands in intact cells are real structures and it is more safe to observe cells pretreated with detergents in embedment-free sections.
