In order to investigate the functional relationship between the astrocyte and capillary Ca^<2+>-ATPase, the so-called cold lesion was produced by dry ice to exposed rat skulls, and this enzyme activity and the morphological change of the astrocyte were studied electron-microscopically and ultracytochemically by using the method of Ando et al. Under the normal condition, the cytochemical reaction (CR) for Ca^<2+>-ATPase was mild. 20 minutes after the cold injury, the astrocytic end-feet along the capillary were found to be swollen, when the CR was observed to be moderate. Deposits of the reaction product (RP) were slightly coarser and appeared to pile up on the abluminal surface. At 4, 7 and 15 days, detachment of the astrocytic end-feet from the capillary wall was observed and the CR was intense especially on the abluminal surface of the uncovered capillary. Capillary Ca^<2+>-ATPase activity increased as the brain edema progressed. And it could be possible that this enzyme was closely related to the BBB. At 2 months, reactive astrocytes recovered surviving capillaries. The CR was mild and deposits were coarser, the number decreasing on both surfaces. It was apparent that the nature and localization of the deposits in the lesion repair were different from those in the normal condition. This might be due to the functional differences between the normal and reactive astrocytes for the BBB. In other words, in this study, RP of mild intensity was observed on the CEC with the astrocytic processes along the capillary wall and that the CEC showed intense RP without astrocytic processes on the capillary wall.
