The Triton X-100 soluble nonspecific cross-reacting antigens (NCAs) recognized by MAb 3H3 have apparent mol. wts of 55,000 (NCA-55) in a pancreatic cancer cell line and 90,000 (NCA-90) in granulocytes using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). It was reported that, when granulocytes and epitherial cells in lung and pancreas were extracted with perchloric acid, mol. wts of 55,000 NCA existed in all of them, and mol. wet of 95,000 NCA existed only in granulocytes. But using Triton X-100 as detergent for the extraction, there have been almost no reports about this. In this paper, NCAs extracted by Triton X-100 were characterized by SDS-PAGE, western blotting, 2 dimensional electrophoresis using MAb 3H3. And the conclusions are summurized as follows: 1) When extracting NCAs using not perchloric acid but detergents, several NCAs exist in the extract. NCA-55 and NCA-90 are also extracted by the method. 2) NCA-55 exists in both perchloric acid and Triton X-100 treatment, so NCA-55 is not degradation product by perchloric acid treatment. 3) In a pancreatic cancer cell line (BxPC-3), NCA-55 is well produced with CEA, but NCA-90 is not produced. Although NCAs in granulocyte are consist of NCA-90 and NCA-55, NCA-55 exists less than NCA-90 in granulocyte. 4) NCA-55 from the pancreatic cancer cell line has chromatographically polymorphism. NCA-55 is consist of nearby four isoelectic points (pH 4.8, 5.3, 5.6, 5.8) 55 kD proteins, and pH 5.8 NCA-55 may more exist than others.
