Mononuclear cells obtained from bone marrow or peripheral blood cells of 22 patients with acute myeloid leukemia were incubated in liquid culture system with and without CSFs. In two of 22 cases, leukemic cells proliferated wihout CSFs. G-CSF stimulated the proliferation of leukemic cells from eight of 22 patients and GM-CSF did from seven of fifteen patients. This stimulation by CSFs appeared to be dose-dependent. In addition, additive and synergistic effects were observed in four of eleven leukemic cells stimulated with combined G-CSF and GM-CSF in vitro. Only one case showed differentiation with G-CSF. Out of nine patients given G-CSF clinically, three showed in vitro growth of their leukemic cells. In one patient with a striking cell proliferation by both CSFs in vitro, his leukemic cells incresed in number after the administration of G-CSF (iv). These observations suggest that leukemic clone may have some sensitivity to CSFs in vitro and in vivo.
