The apparently increased neurotrophic activities in AD brain has been shown to be a result of loss of grouth inhibitory factor (GIF), which is present in normal adult brain. In order to study the altered gene expressions of the GIF in AD brains, a fundamental step is to isolate intact messenger RNAs (mRNAs) from postmortem human brains. We studied stabilities of mRNAs in postmortem human brain and showed intact mRNAs expressed in neurons, astrocytes, or oligodendrocytes can be isolated from postmortem brains for up to 12 hours after death. In AD brains which were pathologically confirmed, the expression of the GIF mRNA was investigated by Northern blotting.
