The following results were obtained:(1) Acute administration of phencyclidine (PCP) to rats induced a complex behavioral pattern which consisted of high locomotor activity, stereotyped behavior and ataxia. After 10 days of chronic PCP administration, PCP-induced stereotypy and ataxia decreased but the locomotor activity increased; (2) In frontal cortex (Fc), NTLI decreased significantly in both the acute and chronic PCP groups; (3) Chronic administration of methamphetamine (MAP) to rats produced progressive augmentation in response, characterized by a more rapid onset and an increased magnitude of stereotypy; (4) After a dose of 10mg/kg/day of MAP had been given for 5, 10 and 20 days, NTLI in limbic forebrain (LF) significantly increased from 14.6pg/mg wet weight (control) to 21.2 (5 days), 23.7 (10 days) and 22.8 (20 days). Similar results were also observed in caudate putamen (CP) and midbrain (MB); (5) When rats were administered MAP (50mg/kg) and sacrificed after 1, 6, 12, 24, 36 and 48h, the increase in NTLI in LF became evident first at 12h and persisted through 24h. NTLI in the CP began to increase at 6h after the MAP injection. This increase persisted for at least an additional 30h. In the MB, NTLI significantly increased from 24 to 36h after MAP administration; (6) The increase of NTLI in LF, CP and MB were dependent on the dose of MAP.
