The endothelial cells were incubated with various concentrations of free AA, 3, 15, 30, 150, 300μM, dissolved in minimal essential medium. The endothelial cell damage was estimated by ^<51>Cr cytotoxicity test and the ability to produce prostacyclin by the endothelial cells after the incubation with free AA. ^<51>Cr release from endothelial cells increased time dependently after the incubation with ≧15μM of free AA, and reached maximum at the end of 24 hours incubation. Pretreatment with dexamethasone, a phospholipase A_2 inhibitor, partially inhibited ^<51>Cr release. However indomethacin, a cyclooxygenase inhibitor and BW755C, a cyclooxygenase and lipoxygenase inhibitor, had no effect on it. Prostacyclin producton by endothelial cells also decreased time dependently after the incubation with ≧30μM of free AA. These findings suggested that free AA itself, not the metabolites, induced the endothelial cell damage.
