PBLs collected from 21 maligmant brain tumor patients were examined by two-color flow cytometric analysis using Leu series monoclonal antibodies. Patients were divided into three groups according to the treatment regimen; (R) brain irradiation only, n=4; (A) (R)+1-(4-amino-2-methyl-5-pyrimidyl) methyl-3-nitrosourea hydrochloride, n=7; and (I) (A)+OK 432 or interferon-β, n=10. Results presented herein are summarized as below; 1) T(Leu 4+12-) cells have increased in all groups and B (Leu 4-12+) cells have decreased significantly (p<0.05) in groups R and A. B cells have also decreased in group I. 2) All groups showed an increase in activated T (Leu 4+HLADR+) cells before the treatment. Moreover, in groups R and A, these activated T cells have increased significantly (p<0.05) during the therapy. Activated T cells have also increased in group I. 3) CD4/CD8 ratios have decreased in groups R and A. In group I, however, it has remained nearly constant during the therapy. 4) Cytotoxic T (Leu2a+15-) cells have increased almost in parallel with activated T cells. So, the major component of the increased aetivated Tcells were thought to be the cytotoxic Tcells. 5) Strong NK(Leu 7-11+) cells have increased in all groups and moderate NK (Leu7+11+) cells have increased in groups R and A.
