To better understand the mechanism of neuronal death in Alzheimer's disease, we have undertaken molecular cloning of a cDNA for human growth inhibitory factor (GIF), which has recently been identified as a unique protein with malkedly reduced expression in Alzheimer's disease. Molecular cloning of cDNA for human GIF revealed that the GIF cDNA has a strikingly high homology to human metallothioneins. The tissue distribution of the GIF mRNA expression is exclusively specific to the central nervous system, indicating that the trasncriptional regulation of GIF gene is quite different from those of metallothionein genes despite of it high homology. Furthermore, it was demonstrated that the GIF mRNA expression is markedly reduced in Alzheimer's disease.
