Tracheal cryopreservation was done by a programed freezing process. After achieving the temperature, trachea was transferred into liquid nitrogen and preserved for 30 days. Light microscopic and scanning electron microscopic findings revealed epithelial desquamation after cryopreservation. In fresh trachea positive reactions could be detected in the epithelium by immunostaining with the monoclonal antibody (mAb) OX6 (anti-MHC class II antigen), but in cryopreserved trachea positive cells could not be detected because of epithelial desquamation. The experimental design was orthotopic tracheal transplantation in rat. After transplantation antibiotics and immunosuppresive angents were not administrated. Three groups were designed:Group A animals (n=13) received fresh autograft;Group B animals (n=16) received cryopreserved allograft;Group C animals (n=17) received fresh allograft. Infiltration of mononuclear cells was revealed to be more prominent in Group C (fresh allograft) than in Group B (cryopreserved allograft).
