In this study, the part of this mechanism was examined by molecular technique. Heterotopic heart and orthtopic left lung transplantation were performed. Experimental groups were divided as follows:Group 1, control group which were injected control antibody once a day for two consecutive days, beginning two days before transplantation. Group 2, preoperative treatment group which were injected R73 for two days as group 1. Group 3, postoperative treatment group which were injected R73 once a day for seven consecutive days, beginning on the day of transplantation. Heart grafts survived more than 200 days only in group 2. On the other hand, lung grafts were rejected with five days delay compared with control group. Phenotypic and functional analyses of T cells obtained from rats treated with R73 suggest that TCR molecules and function of T cells are transiently down-regulated at the time of transplantation. In RNase protection assay, expression of cytokine mRNA on grafts was suppressed only in group 2. These data suggest that exposure to alloantigen during T cell modulation suppress cytokine production which initiate and enhance graft rejection. In normal lung but not heart, cytokine and adhesion molecule were detected in mRNA level.
