Previous CSD induced tolerance against subsequent ischemic insult. Ketamine, an NMDA antagonist and proven inhibitor of CSD, abolished this preconditioning effect. Volatile anesthetics, halothane, isoflurane and sevoflurane induced suppression of CSD elicitation in a dose dependent manner. With increasing concentrations of each volatile anesthetic, there was dose-related reduction in CSD frequency but not in CSD amplitude. However, volatile anesthetics isoflurane and sevoflurane did not have any effect on the signal transduction pathways that lead to c-fos expression. The differences in propagation of membrane depolarization between neurons that had acquired ischemic tolerance and neurons that had not acquired it were also investigated. In comparison to control slices, propagation was significantly depressed and depolarization was shortened in slices from gerbils subjected to 5 min of ischemia. Slices from gerbils who had acquired ischemic tolerance demonstrated only a slight reduction in propagation.
