PAI-1 mRNA was expressed in the whole eye, lung, myocardium, aorta, and kidney in normal rats while it was not detected or under detection level in the whole brain, liver and skeletal muscle. In diabetic rats, the level of PAI-1 mRNA increased in all the organs expressing PAI-1 mRNA except for the kidney. t-PA mRNA was expressed in the whole brain, whole eye, lung, myocardium, and kidney and, in diabetic rats, its level was found to be increased except for that in the kidney. The expression pattern in systemic organs of normal rats and its change in diabetic rats was quite similar between PAI-1 and t-PA mRNAs, suggesting their implications in the development of microangiopathy characteristic of diabetes mellitus. PAI-1 and t-PA mRNA expression in the kidney was found to be highly enriched in the glomeruli of both normal and diabetic rats. Double immunofluorescence staining with monoclonal antibody (mAb 5-1-6) against slit diaphragm revealed the co-localization of anti-PAI-1 antibody and mAb 5-1-6.
