{"created":"2021-03-01T06:15:18.938885+00:00","id":11131,"links":{},"metadata":{"_buckets":{"deposit":"6b3da9d0-39b1-478e-9fd7-38567800e789"},"_deposit":{"id":"11131","owners":[],"pid":{"revision_id":0,"type":"depid","value":"11131"},"status":"published"},"_oai":{"id":"oai:niigata-u.repo.nii.ac.jp:00011131","sets":["453:456","471:537:538:989"]},"item_7_alternative_title_1":{"attribute_name":"その他のタイトル","attribute_value_mlt":[{"subitem_alternative_title":"Enhanced Neovascularization and Tissue Regeneration in a Biodegradable Vascular Graft by Implantation of Autologous Bone Marrow Mononuclear Cells"}]},"item_7_biblio_info_6":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2008-07","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"7","bibliographicPageEnd":"381","bibliographicPageStart":"372","bibliographicVolumeNumber":"122","bibliographic_titles":[{"bibliographic_title":"新潟医学会雑誌"},{"bibliographic_title":"新潟医学会雑誌","bibliographic_titleLang":"en"}]}]},"item_7_description_4":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"【背景と日的】生体吸収性材料と自己細胞を用いたTissue Engineering技術による再生医療が臨床応用され始めている. 移植されたグラフト内への毛細血管の新生により, 組織再生が促されると考えられる. さまざまな種類の細胞に分化する可能性をもつ未分化な細胞を多く含む骨髄単核球細胞(BMC)を生体吸収性ポリマーに播種することにより毛細血管新生が促進されると考えられるため, ラット右室流出路再建モデルを用いてBMC播種の効果を検討した.【方法】播種する細胞には, ラット大腿骨および脛骨より採取したBMCを用いた. 生体吸収性材料としてポリカプロラクトン(PCL)とポリ乳酸(PLA)(50:50)の共重合体(P(CL/LA))ポリマーを使用した. 細胞を播種しないポリマー(PCLA群)と1×10^5/mm^2個のBMCを播種したポリマー(BMC郡)を用いて, ラット右室流出路に作成した貫壁性の欠損を再建した. 2週, 4週および8週後(各期間, 各群n=6)の病理組織を血管新生および組織再生について検討した. 【結果】von Willebrand factorによる免疫組織学的染色では, 2週後で両群ともに心腔面の内皮化を認めた. 移植したポリマー内の新生血管数は2週後(PCLA: 85.7± 17.6/mm^2, BMC: 149.8± 16.1/mm^2, p<0.05)ではBMC群が有意に多く, 4週後(PCLA: 210.5± 20.1/mm^2, BMC: 347.0± 78.0/mm^2, p=0.121)ではBMC群が多い傾向があった. また, BMC群ではα-SMA陽生の血管平滑筋細胞に取り囲まれた成熟した血管も多く認めた.
