Although the pathophysiology of the formation of senile pleque, a cardinal pathological feature of Alzheimer's disease, is becomming well understood, the mechanisms of neuronal degeneration are still unclear. To better understand the mechanism of neuronal death in Alzheimer's disease, we have undertaken molecular cloning of a cDNA for human growth inhibitory factor (GIF), which has recently been identified as a unique protein with malkedly reduced expression in Alzheimer's disease. Molecular cloning of cDNA for human GIF revealed that the GIF cDNA has a strikingly high homology to human metallothioneins. The tissue distribution of the GIF mRNA expression is exclusively specific to the central nervous system, indicating that the trasncriptional regulation of GIF gene is quite different from those of metallothionein genes despite of it high homology. Furthermore, it was demonstrated that the GIF mRNA expression is markedly reduced in Alzheimer's disease. The data indicate that it is crucially important to elucidate the mechanism of the downregulation of the GIF gene in Alzheimer's disease.
