In Order to elucidate the implication of HLA-class II antigens in the genesis of unexplained recurrent abortion, the frequency of HLA-DRB 1 , -DQB 1 , and -DPB 1 genotypes in patients with unexplained recurrent abortion and the compatibility of the genotypes between patient couples were studied by using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Twenty-eight patients with unexplained primary recurrent abortion and their husbands were typed for HLA-DRB 1 , -DQB 1 , and -DPB 1 genotypes. Polymorphic regions at the second exon of HLA-DRB 1 , -DQB 1 , or -DPB 1 were selectively amplified by using the PCR-primers. After amplification, DNAs were digested with restriction endonucleases, and subjected to electrophoresis in a 12% polyacrilamide gel to determine each HLA genotypes. In this study, the frequency of some genotypes was significantly higher in patients group compared with normal population, although no significant compatibility of HLA genotypes could be observed between patient couples compared with normal fertile couples. These data may represent a new class II association with the genesis of unexplained recurrent abortion.
