The localization of carcinoembryonic antigen (CEA) in the eccrine gland apparatus of normal skin and eccrine gland tumors was reevaluated using anti-CEA monoclonal antibodies (MoAbs) which are more specific to colonic CEA. The staining patterns were compared with those of anti-CEA polyclonal antibody (PoAb). The upper portion of the eccrine duct was reactive with all the MoAbs and PoAb. However, the lower duct and secretory portion was weakly or negatively reactive with these MoAbs, while PoAb reacted strongly with these areas. The staining patterns in the eccrine gland tumors implied the direction of differentiation of the tumor cells. The tumor cells which differentiate to the upper eccrine duct showed positive reactivity with MoAbs and PoAb. Those differentiating towards the secretory portion or lower duct reacted strongly with PoAb but weakly or negatively with MoAbs. These results suggest that the antigen which has been regarded as CEA in the skin is not identical to colonic CEA, and shows heterogeneity. Therefore, the presence of some CEA-related antigens was proposed.
