Changes in arachidonic acid metabolism in human leukemia cells during differerntiation are poorly defined. In this study arachidonic acid metabolites produced by eight different types of leukemia cell lines (HL-60, K-562, KG-1, KU-812, Molt-3, HEL, U-937 and J-111) before and after stimulation with phorbol 12-myristate 13-acetate (PMA) or 1-α, 25-dihydroxy vitamin D_3 in vitro were analyzed using high performance liquid chromatography (HPLC). Some myeloid cell lines (HL-60, K-562, KU-812, J-111 and KG-1) produced PGE_2 through the metabolic pathway of cyclooxygenase by stimulation with arachidonic acid even without differentiation-inducing substances. Interestingly, HL-60 cell line produced high levels of not only PGE_2 but also metabolites such as PGD_2 and TXB_2, suggesting the presence of metabolic pathway of cyclooxygenase which may differ other cell lines. In HL-60, K-562 and KG-1, which are myeloid cell lines, and in J-111, a monocytic leukemia cell line, the production of PGE_2 was increased by treatment with PMA, and morphologically monocyte-like change were observed. On the other hand, treatment with 1-α, 25-dihydroxy vitamin D_3, which has been reported to induce differentiation into granulocytes, caused no increase in PGE_2 production. These results suggest that the cells which differentiate into monocyte-macrophages mainly produce PGE_2 through the metabolic pathway of cyclooxygenase, and their differentiation into granulocytes or lymphocytes is not mediated by cyclooxygenase.
