By molecular biology new important proteins and peptides have been cloned. Detection, localization and quantitation of these proteines and peptides in cells, tissues or organs are aimed in cell biology or pathology. Immunological techniques using antibodies as probes have been employed for this purpose, however, antibodies against the newly-discovered substances are often unavailable. To solve the problem, methods to detect mRNA using nucleic acids as probes are recently devised. This review introduces two representative techniques to detect mRNA in total cellular RNA extracted from cell or tissue samples and to localize mRNA in tissue sections. Ribonuclease (RNase) protection assay is more simple, sensitive and quantitative than Northern blot analysis for detection of mRNA in total cellular RNA samples. In addition, expression of several different mRNAs can be examined in a single sample by this method using multiple probes. In situ hybridization is a method to localize mRNA in cells in tissue sections. As their applications presented are expression of mRNA for cytokines and growth factors in glomeruli of rats with experimental glomerulonephritis and localization of mRNA for mRNA for type A atrial natriuretic peptide receptor in glomeruli.
