We observed the histological features of the repair process in a destructive lesion of the rat brain during the proliferation of neuroepithelial cells (NECs). Pregnant rats on day 12 of gestation were given a single intravenous injection of 40 mg/kg ethylnitrosourea (ENU), and the cytotoxic effects on the brains of the embryos (E12) and the subsequent repair process were studied chronologically. The results are summarized as follows. 1) There hours after ENU administration, shrunken and darkened degenerating cells appeared in the NEC layer. 2) NECs engulfed and digested these degenerating cells. 3) These NECs still appeared to perform elevator movement and had the potential to proliferate. 4) The width of the NEC layer diminished after administration of ENU, reached a minimum at 6h, and recovered rapidly by 10h. On E14, there was no significant difference in the width between the ENU-treated rats and the untreated controls. 5) On postnatal day 21, there was no significant difference between the ENU-treated rats and the controls in gross appearance and brain weight. Histologically, the former showed, no distinct changes, including neuron loss or gliosis, compared with the latter. In this experiment, a single intravenous injection of ENU caused degeneration and death of the NECs. On postnatal day 21, we were unable to find any significant differences between the treated rats and the controls. We consider that in the early stage of brain developments, NECs participate actively in the process of brain lesion repair ; eliminating the degenerating NECs and having the potential to compensate for the loss.
