Following peripheral nerve lesions in the adult, Aβ fiber sprouts from deeper laminae into lamina II (substantia gelatinosa) which is thought to be involved in nociceptive processing. However, it is not clear whether Aβ fibers transmit the sensory information to the new territory after peripheral nerve transection. In the present study we found both monosynaptic and polysynaptic responses elicited by Aβ fiber in lamina II neurons of the adult rat with sciatic nerve transection. These observations suggest that sensory information conveyed by Aβ afferents is directly or indirectly transmitted to lamina II neurons which hardly receive Aβ afferent inputs in normal condition. This reorganization of sensory circuitry in the spinal dorsal horn may be responsible for the pathological pain such as allodynia.
