Syntaxin 1A is a key component of the SNARE mechanism. Since exocytosis requires both Ca^\u003c2+\u003e and ATP, we searched for Ca^\u003c2+\u003e/ATP-dependent syntaxin-binding proteins from the rat brain and discovered CaMK II At Ca^\u003c2+\u003e concentrations exceeding 10^\u003c-6\u003eM, only autophosphorylated CaMK II bound to syntaxin. CaMK II bound to the linker domain of syntaxin, unlike any other known syntaxin-binding proteins. CaMK II-syntaxin complexes were also detected in synaptosomes by immunoprecipitation, and when reconstituted in vitro, recruited larger amounts of synaptotagmin and SNAP-25 than syntaxin alone. The microinjected CaMK II-binding domain of syntaxin specifically affected exocytosis in chromaffin cells and in neurons. 内容記述 | In neurons, exocytosis is performed through the SNARE mechanism. Syntaxin 1A is a key component of the SNARE mechanism. Since exocytosis requires both Ca^<2+> and ATP, we searched for Ca^<2+>/ATP-dependent syntaxin-binding proteins from the rat brain and discovered CaMK II At Ca^<2+> concentrations exceeding 10^<-6>M, only autophosphorylated CaMK II bound to syntaxin. CaMK II bound to the linker domain of syntaxin, unlike any other known syntaxin-binding proteins. CaMK II-syntaxin complexes were also detected in synaptosomes by immunoprecipitation, and when reconstituted in vitro, recruited larger amounts of synaptotagmin and SNAP-25 than syntaxin alone. The microinjected CaMK II-binding domain of syntaxin specifically affected exocytosis in chromaffin cells and in neurons. These results indicate that the Ca^<2+>/ATP-dependent binding of CaMK II to syntaxin is an important process in the regulation of exocytosis. | |||||
